Anticholesterol Bioactive Peptides from Bromelain Hydrolysates of Mangrove Sonneratia alba Protein

Cholesterol with high levels in blood vessels can cause atherosclerosis, stroke, and sudden heart attack. Isolates soy protein have bioactive peptides that the potential as anticholesterol. This research aims to determine optimum conditions of hydrolysis characterize from isolate hydrolysates by bromelain enzyme. Hydrolysis optimization conducted using enzyme 0,2%; 0,5% (w/v) variation incubation time 0, 1, 2, 3, 4, 5 6 hours at 45, 50 55°C. Protein analyzed degree (% DH) tested anticholesterol activity test through HMG-CoA reductase inhibition pravastatin a positive control. The results showed were obtained 2 hours, temperature 45 °C concentration is DH value 40,22%. highest was hydrolysate percent 82,80% (7.371 ppm). SDS-PAGE analysis show appearance bands under 10 KDa fractionation peptide fragments has molecular weight 2779 2609 Da.
